Job Description Summary The successful candidate willbe accountable for the successful and appropriate in vitro and invivo characterisation of binding specificity of candidate and Tcell activation and T cell-mediated killing of bi-specific TCRmolecules to cells. - In this role you will be accountable for thesuccessful and appropriate in vitro characterisation of bindingspecificity of candidate and T cell activation and T cell mediatedkilling of bi-specific TCR molecules to cells. - This role requiresstrong team leadership, scientific understanding, and expertise inimmunology, T cell assays, cellular biology, and molecular biology.Experience with lentiviral transduction and cloning is desirable. -You will have responsibility for leading the day-to-day running ofthe preclinical biology group. Key Responsibilities - Lead theday-to-day operations of the preclinical biology group. - Developand implement biological assays to ensure candidate soluble TCRstranslate to potent and safe clinical molecules. - Oversee allbiological testing activities related to soluble TCR moleculesgenerated by the Protein Engineering team. - Manage a team of cellbiologists, initially comprising 3-4 individuals, with potentialfor team expansion as candidate targets are nominated. - Determineand implement appropriate in vitro testing strategies for efficacy,safety, and underlying molecular mechanisms of soluble TCRs,including establishing assays and sourcing cell lines and tissuesamples. - In the future there may be the opportunity to implementappropriate in vivo testing strategies, utilising outsourcedresources if required. - Collaborate effectively with theleadership team, including the Head of Protein Engineering and Headof Protein Production, to guide optimization and development ofclinical soluble TCRs. Experience/Qualifications - PhD in arelevant biological subject with at least 5 years of subsequentbiotechnology and/or pharmaceutical industry experience in R&Dteams. - Direct hands-on laboratory experience in testing thesafety and efficacy of soluble high-affinity TCR molecules. - Deepfamiliarity with traditional cell biology and molecular biologytechniques, including cell binding assays, T cell activation, and Tcell killing both in vitro and in vivo. - Experience in generatingisogenic cell lines using lentiviral transduction, growing a largearray of cell lines, human organoids, and primary human cells. -Expertise in complex multi-color flow cytometry, EliSpot assays,luminometry, fluorescence, and in vivo assays of safety andefficacy. - Proven leadership skills, including successfuldevelopment and implementation of scientific strategies, andability to work with other leaders and support staff. - Experiencein managing, mentoring, developing, and motivating teams ofscientists. - Experience in managing budgets, agreeing anddelivering timelines of work, and writing reports. - Ability topresent work to the Senior Leadership Team. - Ability to publishand present work done by your team and in partnership with otherYellowstone teams. - Familiarity with requirements for developmentand submission of preclinical data packages for a Clinical TrialAuthorisation to MHRA and a New Drug Application to the FDA andEMA.
